Description

Description: This assay employs a two-site sandwich ELISA to quantitate FOXP3 in samples. An antibody specific for FOXP3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FOXP3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FOXP3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FOXP3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Foxp3 is a member of the forkhead/winged-helix family of transcriptional regulators and functions as the master regulator in the development and function of regulatory T cells. Human FOXP3 genes contain 11 coding exons. Exon-intron boundaries are identical across the coding regions of the mouse and human genes. By genomic sequence analysis, the FOXP3 gene maps to chromosome Xp11.23.
Foxp3 gene is mutated in the X-linked syndrome of immunodysregulation, polyendocrinopathy, and enteropathy (IPEX). These mutations were in the forkhead domain of FOXP3, indicating that the mutations may disrupt critical DNA interactions
